Diversidad Bioquímica y Molecular en Frutillas Chilenas (Fragaria ChiloensisL. Duch.) y su Implicancia en el Mejoramiento Genético de la Especie

Resumen
La frutilla chilena (
Fragaria chiloensis
L. Duch.) es una especie nativa de Chile que fue domesticada antes de la
llegada de los españoles. Esta especie es uno de los progenitores de
la frutilla cultivada (
F. x ananassa
) y se encuentra distribuida en una amplia zona del país, colonizando
diferentes sistemas agroecológicos. Aparentemente, en estos diversos
hábitats se han desarrollado diferentes ecotipos que presentan una
gran diversidad de características morfológicas y agronómicas.
Los objetivos del trabajo fueron: a) determinar la diversidad genética
de una muestra representativa de frutillas chilenas usando marcadores bioquímicos
y moleculares; b) comparar la información previamente generada por
la amplificación de ADN polimórfico al azar (RAPD) en las mismas
accesiones; y c) determinar el posible uso de esta información en un
programa de mejoramiento de la especie. En este trabajo se analizaron 83
accesiones silvestres con los sistemas enzimáticos fosfoglucosa isomerasa
(GPI), leucina aminopeptidasa (LAP), y fosfoglucomutasa (PGM) y 61 accesiones
mediante la amplificación de fragmentos polimórficos (AFLP). Los
resultados del estudio enzimático indicaron una baja diversidad genética
y sólo algunos genotipos fueron polimórficos para LAP y GPI. Los
resultados de AFLP detectaron también un bajo nivel de polimorfismo
entre las accesiones analizadas. Se discuten estos resultados y sus implicaciones
sobre el mejoramiento genético de la especie.

Biochemical and Molecular Diversity in Chilean Strawberries (Fragaria ChiloensisL. Duch.) and its Implication for Genetic Improvement of the Species

Abstract
The Chilean wild strawberry (
Fragaria chiloensis
L. Duch.) is a native species of Chile that was domesticated before the
arrival of the Spanish. This species is one of the progenitors of the commercial
strawberry (
F. x ananassa
), and is widely distributed throughout the country in variable agroecological
systems. Apparently, these diverse habitats have forced this species to
develop a high variability of morphological and agronomic traits. The objectives
of this work were: a) to determine the genetic diversity of a representative
sample of the Chilean strawberry using biochemical and molecular markers;
b) to compare this information by random amplified polymorphic DNA (RAPD)
diversity within the same accessions; and c) to determine the feasibility
of using this information for a breeding improvement program. Eighty-three
Chilean wild accessions were analyzed by three isozyme systems, glucose
phosphate isomerase (GPI), leucine amino peptidase (LAP) and phosphoglucomutase
(PGM), and sixty-one of them were analyzed by amplified fragment length
polymorphism (AFLP). The results indicated a low level of genetic diversity,
only a few accessions were polymorphic for LAP and GPI. The AFLP analysis
also detected a low level of polymorphism among the accessions evaluated.
The results and their implications for future genetic improvement of the
species are discussed.
