Abstract

Yeast derived recombinant HBsAg is lyophilized as a preservation method for long term storage. Three different lyophilization cycles were assayed. The freezing temperature was -50 °C, the rate of ice sublimation was 10 °C/h and also sublimation at 10 °C and 20 °C was tested with temperature for desorption of residual water of 25 °C and 30 °C respectively. Additives like dextran, maltose, sucrose, magnesium chloride and trehalose were studied in the lyophilization and for all of them, the HBsAg activity recovery after lyophilization was about 100%. Residual humidity of lyophilized samples was of 2% in all the cases. The structure of assembled particles of HBsAg was not affected by lyophilization pr Cess using sucrose and maltose as additives.

 es Influencia del uso de aditivos en la liofilizacion del antigeno de superficie recombinante del virus de la Hepatitis B

Resumen

El antigeno de superficie del virus de la hepatitis B es liofilizado como m etodo de conservaci on. Se estudiaron tres ciclos de liofilizaci on, la temperatura de congelacion fue -50 °C en todos los casos; se estudi o un esquema lento de sublimaci on del hielo a raz on de 10 °C/h, se ensay o la sublimaci on a -10 °C y 20 °C; las temperaturas de desorci on del agua residual fueron 25 °C y 30 °C respectivamente. Se realiz o un estudio de liofilizaci on empleando aditivos como dextrana, maltosa, sacarosa, cloruro de magnesio y trehalosa; los valores de actividad del liofilizado no mostraron diferencias respecto a los valores iniciales; el porcentaje de humedad residual de los liofilizados fue menor del 2%; se obtuvo una correcta particulaci on del antigeno determinado por cromatograf ia de esclusion molecular por HPLC al emplear bajas concentraciones de sacarosa y maltosa.
