Abstract

This report shows the stability test studies under accelerated and normal storage conditions carried out with recombinant human alpha 2b interferon (hu-r alpha 2b IFN) in phosphate buffer 0.1M, pH 7.0, with and without albumin, in order to establish its shelf-life at refrigerating and frozen condiions. According to the accelerated study we concluded that no alterations will interfere the recognition of hu-r alpha 2b IFN in ELISA in at least five years when stored at -70 or -20 C. Otherwise, when stored at 4 C, a loss of a 10 % may occur in one year. We corroborated this when the presence of new structures which might affect the protein immunological recognition were detected by RP-HPLC. No stabilizing properties of albumin on hu-r alpha 2b IFN were observed at least when it is in phosphate buffer 0.1M, pH 7.0 and under accelerated storing conditions.

 es Tiempo de vida del interferon alfa 2b humano recombinante en diferentes condiciones de almacenamiento

Resumen

Este trabajo presenta un estudio de estabilidad acelerada realizado al interferon alfa 2b humano recombinante (IFN alfa 2b hu-r) en tampon fosfato 0,1M, pH 7,0 bajo condiciones reales de almacenamiento para determinar su tiempo de durabilidad en condiciones de refrigeracion y de congelacion. Se probaron dos variantes de almacenaje: en presencia o en ausencia de albumina humana. Del estudio acelerado se obtuvo que en el ensayo ELISA despues de cinco anos almacenado a las temperaturas de -20 o -70 C no ocurren alteraciones que impidan el reconocimiento del IFN alfa 2b hu-r. Si se almacena a 4 C la perdida del 10 % puede ocurrir al cabo de un ano aproximadamente. Esto se corroboro por almacenamiento a 4 C en tiempo real monitoreado por HPLC-RP donde se observo que al ano aparecen nuevas estructuras que pueden interferir en el reconocimiento. Los resultados mostraron que la albumina no posee propiedades estabilizantes sobre el IFN alfa 2b hu-r al menos cuando este se encuentra en tampon fosfato 0,1M, pH 7,0 y bajo condiciones aceleradas de almacenamiento.
