Resumen

En muestras de individuos sanos y enfermos tratados o no con interferón (IFN) alfa-2b recombinante (Heberón alfa R) o leucocitario natural (Heberón alfa N) se estudió la presencia de anticuerpos anti-IFN mediante un ensayo inmunoenzimático (ELISA) tipo "sandwich" (IFN alfa-2b/muestra/proteína A-peroxidasa) con confirmación de la especificidad. También se investigó la neutralización de la actividad antiviral de IFN alfa-2b. Por ELISA no se encontró anticuerpos anti-IFN en 184 individuos no tratados mientras que en otras muestras evaluadas por bioensayo hubo actividad neutralizante en sujetos con hepatitis viral crónica (8/181; 4,4 %) y neoplasias (11/121; 9,1 %) y no en otros 98 con otras enfermedades o sanos. No se detectó anticuerpos anti-IFN en ninguno de los individuos tratados con IFN leucocitario, 27 de ellos evaluados por ELISA y 52 por neutralización de la actividad antiviral. Después del tratamiento con IFN alfa-2b se confirmó mediante ELISA la positividad en una niña con linfangioma y en un adulto con papilomatosis laríngea entre 177 pacientes. Los sueros positivos por ELISA tuvieron títulos neutralizantes de 1:25 y 1:64 000, respectivamente. El tratamiento indujo actividad neutralizante en 13,1 % (19/145) de los pacientes evaluados por bioensayo. La incidencia total de anticuerpos inducidos por el tratamiento, evaluada por cualquiera de los métodos, fue 6,4 % (19/296). Esta cifra es similar a la reportada para el IFN alfa-2b.

Abstract

We studied the presence of anti-interferon antibodies in samples of healthy or sick individuals treated or not with natural leukocyte (Heberón alfa N) or recombinant alpha-2b (Heberón alfa R) interferon (IFN) by means of an immunoenzymatic assay (ELISA), "sandwich"-type (IFN alpha-2b/sample /protein A-peroxidase) with specificity confirmation. The IFN alpha-2b antiviral activity neutralization was also investigated. No anti-IFN antibodies were found by ELISA in 184 not treated individuals while among other samples the bioassay detected neutralizing activity in subjects with viral chronic hepatitis (8/181; 4.4 %) and neoplasia (11/121; 9.1 %). Ninety-eight healthy subjects or patients with other illness were negative. There was no positivity in leukocyte IFN-treated individuals, 27 of them evaluated by ELISA and 52 by the bioassay. After the treatment with IFN alpha-2b positivity by ELISA was confirmed in a girl with lymphangioma and in an adult with laryngeal papillomatosis among 177 patients. These ELISA-positive sera had neutralizing titers of 1:25 and 1:64 000 respectively. IFN alpha-2b treatment induced neutralizing activity in 13.1 % (19/145) of the patients evaluated by bioassay. The total incidence of treatment-induced antibodies, evaluated by either method, was 6.4 % (19/296). This is similar to what has been reported for IFN alpha-2b.
