Validación de un ELISA para la
cuantificación de IgG humana anti-polisacarido capsular de
Neisseria meningitidis serogrupo C

ABSTRACT
An indirect ELISA against capsular polysaccharide of
Neisseria meningitidis serogroup C was developed to evaluate
the immune response to this component of VA-MENGOC-BC® Cuban
vaccine. Polystyrene plates coated with poly-L-lysine and
polysaccharide antigen ("Finlay" Institute), are first incubated
with human serum samples. Antibodies are detected by addition of
anti-human IgG/alkaline phosphatase conjugate, which recognizes
antibodies against polysaccharide C. The enzymatic reaction is
evidenced by processing of the specific substrate
p-nitrophenylphosfate. The detection limit of the assay is
367 U/mL of specific IgG in human serum. Intra- and interassay lack
of precision was below 10%. Parallelism, linearity and recovery
were 10% of the expected values. The relation between the
standard of the "Finlay" Institute and the PB2 standard from the
Center for Disease Control (CDC), Atlanta, USA, was determined. The
regression equation was CDC = 0.5398 Finlay +143.

RESUMEN
Se desarrolló un ELISA indirecto
utilizando un doble recubrimiento con poli-L-lisina y el
polisacárido capsular de
Neisseria meningitidis
serogrupo C (Instituto "Finlay", Habana, Cuba) para evaluar la
respuesta inmune contra este componente de la vacuna cubana
VA-MENGOC-BC®. Como conjugado, se utilizó anti-IgG
humana/fosfatasa alcalina el cual se une a los anticuerpos
anti-polisacáridos C producidos en humanos, reacción
evidenciada por la degradación del sustrato
específico (
p-nitrofenilfosfato). La detectabilidad
del ensayo fue de 367 U/mL. La imprecisión (intraensayo,
interensayo y total), así como las desviaciones de la
recuperación, linealidad y paralelismo, fueron inferiores a
10%. Se calculó la ecuación de regresión entre
el estándar PB2 del Centro para el Control de Enfermedades
(CDC) de Atlanta, Estados Unidos y el del Instituto "Finlay",
(CDC = 0,5398 Finlay +143), lo que permitió
establecer una relación entre ellos.
