Resumen

El cultivo del maíz en Venezuela es frecuentemente afectado por un grupo de enfermedades foliares, entre las cuales se encuentra la mancha blanca causada por el hongo Curvularia pallescens check for this species in other resources . Esta enfermedad aparece en el período reproductivo de la planta, afectando de manera significativa el rendimiento en granos. Con la finalidad de identificar marcadores SSR, o simple sequence repeat por sus siglas en inglés, asociados con la resistencia a la mancha blanca causada por C. pallescens en maíz, se realizó un análisis de segregantes agrupados o Bulked Segregant Analysis, BSA por su siglas en inglés, asistido con marcadores SSR. Para ello se utilizó una población F2 integrada por 100 plantas derivadas de un cruce entre dos líneas elites (resistente x susceptible), previamente inoculadas con aislamientos puros de C. pallescens. Para el análisis BSA se formaron dos grupos contrastantes (resistentes y susceptibles), los cuales fueron evaluados junto a los dos progenitores con 50 loci SSR. De estos loci solo cuatro resultaron polimórficos, tres de los 46 restantes no amplificaron y 43 de ellos no presentaron bandas diferenciales. Para determinar la asociación entre los loci polimórficos y la resistencia, se empleó una prueba basada en el estadístico chi-cuadrado de independencia (p=0,05), encontrándose asociación entre el locus Bnlg439 y la resistencia a C. pallescens. Adicionalmente, a partir de los resultados de los análisis estadísticos, se calcularon los valores predictivos, especificidad y sensibilidad de los cinco loci polimórficos, entre los cuales el marcador Bnlg439 presentó el valor más alto de especificidad (76%), y el mayor valor predictivo positivo (85%).

Abstract

The Maize crop in Venezuela is frequently affected by a group of foliar diseases, among which one finds the white spot caused by the fungus Curvularia pallescens check for this species in other resources . This disease appears in the plant reproductive stage, affecting grain yield considerably. With the purpose of identifying SSR (simple sequence repeat) markers associated with resistance to white spot in maize, we performed a Bulked Segregant Analysis (BSA) assisted with SSR markers. We used an F2 population comprised by 100 plants, derived from a cross between two lines (resistant x susceptible), which were previously inoculated with pure isolates of C. pallescens. Using the BSA methodology two contrasting groups (resistant and susceptible) were formed, which were evaluated along with their parents with 50 pairs of SSR loci. Only four of the 50 loci evaluated resulted polymorphic, three of the remaining 45 did not amplified, and 43 of them did not show differential bands with the parents or the contrasting groups. To determine the association between the polymorphic loci and resistance, a Chi-square test of independence was applied (p=0.05) finding Bnlg439 locus associated with resistance to C. pallescens. Additionally, from the results of the statistical analyses, we calculated predictive values, specificity and sensibility of the 5 polymorphic loci, among which Bnlg439 marker presented the highest specificity value (76%), and positive predictive value (85%).
