Abstract

The sorghum [ Sorghum bicolor check for this species in other resources (L.) Moench] is the one of the most important food sources for animals, such as pigs and chickens. Due to this aspect it would be important to obtain an effective in vitro regeneration system in order to obtain new cultivars with desirable agronomical characteristics. The aim of this research was to obtain an efficient regeneration system. Mature seeds of the sorghum hybrids Chaguaramas III and VII were cultured in the MS medium supplemented with several hormones (2.4-D, NAA and K, BA or TDZ). Two steps were involved in the regeneration process: I. Callus induction and II. Plant regeneration (in absence of 2.4-D). Calli showing different morphologies were obtained: non embryogenic, embryogenic, with roots, buds or combined of these. The embryogenic frequency was greater with 3 mg.L-1 of 2.4-D + ANA and 1 mg.L-1 of K, whereas the regenerative frequency was greater with BA. When increasing the BA concentration, the organogenesis was favored as indicated by vitrified buds, while the somatic embryogenesis was inhibited. The regeneration occurred by indirect organogenesis or/and somatic embryogenesis, and the origin of the later was unicellular and multicellular. When using BA and TDZ, plants and buds showed non permanent foliar deformations, as well as albino plants. The induction, regeneration and oxidation process varied according to the cultivar.

Resumen

El sorgo [ Sorghum bicolor check for this species in other resources (L.) Moench] es uno de los cereales más relevantes en la elaboración de alimentos concentrados para cerdos y gallinas. Por ello, es importante obtener nuevas cultivares de características agronómicamente importantes a través de técnicas biotecnológicas, por lo cual se debe contar con un sistema de regeneración in vitro, el cual fue el objetivo de este trabajo. Como explantes se utilizaron semillas maduras de los híbridos de sorgo Chaguaramas III y VII. El medio fue MS, con diferentes hormonas (2,4-D, ANA y K, BA o TDZ). El sistema fue de dos etapas: I. Inducción de callo y II. Regeneración (sin 2,4-D). Se obtuvieron callos morfológicamente distintos: no-embriogénicos, embriogénicos, con raíces, con brotes o combinaciones de estos. La mayor frecuencia embriogénica se encontró en medios con 3 mg.L-1 de 2,4-D + ANA y 1 mg.L-1 de K o BA, mientras que la frecuencia regenerativa fue mayor con BA. Al incrementar la concentración de BA se favoreció la organogénesis en detrimento de la embriogénesis somática, observándose brotes vitrificados. La regeneración fue por vía indirecta: organogénesis y embriogénesis somática de origen unicelular y multicelular. Al utilizar BA y TDZ, se regeneraron plantas y brotes con deformaciones foliares no permanentes, así como plantas albinas. La respuesta fue marcadamente varietal, tanto en la etapa de inducción como de regeneración.
