Resumen

En el presente estudio se caracterizó, mediante electroforesis, la expresión de proteínas y del canal de sodio Nav1.3, en ambas astas dorsales medulares (ADM) de rata, en condiciones controles y de inflamación periférica aguda unilateral, tratada o no con indometacina. En ratas Sprague-Dawley, (n=15), anestesiadas (Tiobarbital) se sumergió la pata posterior izquierda en agua a temperatura ambiente (26°C) por 60s (controles; n=5); 60°C (inflamado; n=5) ° 60°C pretratadas con indometacina (pretratado; n=5). Luego de 15 horas, se determinaron, proteínas totales (Bradford y electroforesis) y proteína Nav1.3 (Western Blot) en ambas ADM. La inflamación incrementó las proteínas totales del ADM ipsilateral (+39.8%) y la redujo en la contralateral (-15%). La indometacina disminuyó la expresión de proteínas en inflamación e inhibió aquellas constitutivas ipsilaterales del grupo control con incremento (+14%) contralateral. El canal Nav1.3 no se expresó luego de 15 horas de inflamación. Estos resultados sugieren la respuesta diferencial de cada ADM ante un estímulo nocivo y la participación constitutiva como inducida de las cicloxigenasas en este proceso de expresión protéica.

Abstract

In the present study, the expression of proteins and sodium channel Na1.3, was characterized by electrophoresis in both spinal dorsal horns (ADM) of rats in control conditions and after an acute unilateral peripheral inflammation, and treated or not with indomethacin. Sprague Dawley rats (n=15) were anesthetized (Tiobarbital) and the left hindpaw was immersed in water at room temperature (26°C) for 60s (control; n=5); at 60°C (inflamed, n=5) or 60°C pretreated with indomethacin (pretreated, n=5).After 15 hours, total proteins (Bradford and electrophoresis) and Nav1.3 channel (Western-blotting) determinations were performed. Inflammation increased the ADM proteins ipsilaterally (+39.8%) and reduced it contralaterally (-15%). Indomethacin decreased expression of proteins in inflammation, inhibiting those constitutively expressed in the control group and increasing (+14%) those contralaterally. The channel Nav1.3 was not expressed after 15 hours of inflammation.The inflammation suppressed the protein expression in the contralateral ADM.Indomethacin reduced the ipsilateral protein expression and increased the contralateral. These results suggest the differential response of each dorsal horn to noxious stimuli.
