Abstract

In spite of several methods of bacterial identification existing today, problems still persist in the practice of routine clinical laboratory, for this reason, we consider differentiating, in liquid culture, some pathogenic bacteria in humans by means of +HNMR spectroscopy . Strains of Klebsiella pneumoniae check for this species in other resources , Staphylococcus aureus check for this species in other resources and Streptococcus agalactiae check for this species in other resources , were isolated from patients with different infectious diseases and identified by conventional methods. Later were they cultivated in broth brain-heart- infusion (BBHI), during 18-24 h to 37 C° in 10% of CO2 and analysed by spectroscopy +HNMR. The spectra were processed using the matNMR v. 2.7 software (MATLAB ®). Signals which by their chemical displacement and/or intensity facilitate the differentiation of the species and its culture medium, are found in the aliphatic region. Conclusion: Despite the large number of signals shown by the different species and the BBHI, Klebsiella pneumoniae, Staphylococcus aureus and Streptococcus agalactiae were differentiated by +HNMR spectroscopy, directly in BBHI.

Resumen

Pese a los variados métodos de identificación bacteriana existentes actualmente, aún persisten problemas en la práctica de rutina del Laboratorio clínico, por tal razón, se planteó diferenciar, en cultivo líquido, por espectroscopia +HRMN, algunas bacterias patógenas para humanos. Las cepas de Klebsiella pneumoniae check for this species in other resources , Staphylococcus aureus check for this species in other resources y Streptococcus agalactiae check for this species in other resources , fueron aisladas de pacientes con diferentes cuadros infecciosos e identificadas por métodos convencionales. Posteriormente se cultivaron en Caldo Infusión–cerebro-corazón (CICC), durante 18-24 h a 37°C en 10% de CO2 y se analizaron por espectroscopia +HRMN. Los espectros fueron procesados a través del programa matNMR v. 2.7 (MATLAB®). Las señales que por su desplazamiento químico y/o intensidad facilitaron la diferenciación de las especies y de su medio de cultivo, se encuentran en la región alifática. Conclusión: No obstante el gran número de señales desplegadas por las distintas especies y por el CICC, Klebsiella pneumoniae, Staphylococcus aureus y Streptococcus agalactiae fueron diferenciadas por espectroscopia +HRMN directamente en CICC.
