Abstract

The main purpose of this research was to evaluate and compare metronidazole and tinidazole efficacy against Trichomonas muris check for this species in other resources in hamsters naturally infected and raised under conventional bioterio conditions. There were selected 40 male hamsters based on positiveness to T. muris infection diagnosed for demonstration of the trofozoites on the direct coprological observation. For this purpose metronidazole and tinidazole were administered daily, initially given in one intake during five days at concentrations of 2.5, 5, 6.25, and 7,5 mg/mL by oral way during five days. Thereafter, a second treatment was imposed with two intakes at 7.5 mg/mL. In the first five days of treatment 1, it was obtained a 20% of parasite elimination using tinidazole at a 6.25 mg/mL concentration. A similar response at was obtained by metranidazole at a concentration of 7.5 mg/mL. Additionally, tinidazole efficacy was 40% at a concentration of 7.5 mg/mL. Regarding treatment 2, it was found 100% of effectiveness for tinidazole at 7.5 mg/mL, whereas effectiveness for metronidazole was 50%, with an effective eradication specter lower than the seven days period. It is evidence of a low efficacy of the drugs against tricomicide activity. It is recommended to realize in vitro studies in order to evidence parasite resistance to synthetic drugs derived from nitroimidazole and to stimulate the study and development of new alternative drugs able to proportionate experimental models useful for research.

Resumen

Esta investigación evaluó la eficacia del metronidazol y tinidazol utilizados para eliminar Trichomonas muris check for this species in other resources en hámsters infectados naturalmente provenientes de un bioterio convencional. Se seleccionaron 40 hámsters machos positivos a Trichomonas muris, diagnosticados por demostración del trofozoito en examen coprológico. Se les administraron los antiparasitarios a concentraciones crecientes de 2,5 5,0 6,25 y 7,5 mg/mL por vía oral en una sola toma diaria por 5 días. Posterior a estas aplicaciones se instauró un segundo tratamiento, cuya dosificación fue 7,5 mg/mL, con dos tomas diarias. Finalmente, al culminar dichos tratamientos se les practicó la necropsia. El resultado del primer tratamiento fue una desparasitación del 20% al 6,25 mg/mL con el uso de tinidazol, obteniendo igual resultado para metronidazol al 7,5 mg/mL. En adición, el tinidazol fue eficaz en 40% al 7,5 mg/ mL. Con respecto al tratamiento 2 se logró el 50% de desparasitación con metronidazol al 7,5 mg/mL y el 100% con tinidazol a la misma concentración. Los datos expuestos de manera descriptiva sugieren una baja actividad de los fármacos utilizados y resistencia del parásito; sin embargo, se recomienda el uso del segundo tratamiento aplicado y el realizar estudios prospectivos in vitro e in vivo e incluir los derivados sintéticos del nitroimidazole para así obtener un esquema de administración eficiente en la eliminación del protozoario.
