ABSTRACT
Some variables of the purification process for recombinant human
gamma interferon (rec-hum g IFN) were studied in
order to search for a more economic process that may increase the
productive capacities of this protein. The reduction of the ionic strength
in the renaturation process allowed a recovery of rec-hum g IFN 1.82 times higher in this step as well as a
higher stability of the protein under this condition. A new matrix,
Fractogel EMD 650 SO3- (S) was evaluated in the ionic exchange
chromatography and the working parameters were established. A better
recovery and reproducibility were achieved with a column charge of 10 mg of
protein per miligrame of resin and stepwise gradient at 0.46 - 0.49 M NaCl
in this step. The use of lower ionic strength (0.01 M) showed no
differences in the results obtained and at lower pH values there was an
increase in the recovery of IFN, until a value close to 50 % for this step.
The use of the more economic ion exchange matrix and the best conditions
found in this study allowed a higher recovery in the purification step and
at the same time to own a final product with higher stability and similar
quality attributes.

RESUMEN
Se realizó un estudio de diferentes variables en el proceso
de purificación del interferón gamma humano recombinante (IFN g hu-r) con vistas a
obtener un proceso de purificación más económico, el
cual posibilite el incremento de las capacidades productivas de esta
proteína. Con la disminución de la fuerza iónica en la
renaturalización de 0,1 M a 0,01 M se logró un aumento del
recobrado en el paso de 1,82 veces y la proteína mostró ser
más soluble bajo estas condiciones. Se evaluó la matriz de
intercambio catiónico Fractogel EMD 650 SO3-
(S) y se establecieron los parámetros de trabajo. Se obtuvo un mejor
recobrado y reproducibilidad a una carga de 10 mg/mL de gel y gradiente
escalonado entre 0,46 y 0,49 M de NaCL. Una fuerza iónica inferior
(0,01 M) no mostró diferencias en los resultados y la
disminución del pH produjo un aumento del recobrado, hasta un valor
cercano al 50 % para este paso. Con la utilización de una matriz de
intercambio iónico menos costosa y el empleo de las mejores
condiciones estudiadas en la etapa de purificación, se logró
incrementar el recobrado de la misma y se obtuvo un producto con una mayor
estabilidad e igual calidad.
